Blacklock studied at the University of Edinburgh, and was licensed to preach in 1739. In 1762, he was ordained parish minister of Kirkcudbright, but due to blindness had to resign and retire on an annuity. He went to Edinburgh and there received as boarders university students and boys attending school. In 1767, he received a D.D. degree from Marischal College, University of Aberdeen. Blacklock was one of the earliest literary friends of poet Robert Burns.
